Cat LRG1 SPARCL Kit – added March 26, 2018
LRG1 (leucine-rich alpha-2-glycoprotein-1) is a positive acute phase protein that is elevated in serum during inflammation, infection and certain cancers (in humans). Human CA 724- SPARCL™ Kit  – added Feb 2, 2018
CA 72-4 is a glycoprotein of ~1000 kDa that is  expressed on the surface of cancer cells. Increased levels of CA 72-4 are found in the serum of some cancer patients. The CA 72-4 SPARCL™ assay (CA72-SP-20) is available for research use. Refer to the SPARCL kits webpage for pricing.

Human CA 19-9 SPARCL™ Kit  – added Jan 31, 2018
CA 19-9 is a carbohydrate antigen (Sialyl-LewisA) found on the MUC1 protein that is expressed and shed from some tumor cells. Increased serum levels of CA 19-9 may be associated with pancreatic and gastrointestinal cancer. The CA 19-9 SPARCL™ assay (CA19-SP-20) is available for research use. Human CYFRA 21-1 SPARCL™ Kit  – added Jan 26, 2018
CYFRA 21-1 is a fragment of cytokeratin 19 produced by proteolysis in malignant tissues. It is used as a serum biomarker for some types of cancer. Our rapid and sensitive SPARCL™ assay for human CYFRA 21-1 (CYFR-SP-20) is available for research use. Refer to the SPARCL kits webpage for pricing. Cat Thymidine Kinase 1 SPARCL™ Kit  – added Jan 23, 2018
A cat TK1 SPARCL™ kit is now available (TK1-SP-8). TK1 has potential as a serum biomarker for lymphoma in cats. In studies at Life Diagnostics, we found serum levels ranging from 0 – 95 ng/ml in samples from cats reported as sick by veterinarians. Our TK1 SPARCL™ assays are significantly faster and easier than ELISA and radiometric TK1 assays. They use a single 30-min incubation and require no wash steps. Dog and Human Thymidine Kinase 1 SPARCL™ Kits  – added Jan 19, 2018
We recently made several mouse monoclonal antibodies for thymidine kinase 1. Using HRP and acridan conjugates of the antibiodies we have developed SPARCL kits for dog (TK1-SP-4) and human TK1 (TK1-SP-20). TK1 is a useful serum biomarker for cancer in humans and dogs. Cat Ceruloplasmin ELISA Kit – added Dec 04, 2017

Cat Ceruloplasmin SPARCL™ Kit – added Nov 28, 2017
Ceruloplasmin is a copper transport protein that behaves as a positive acute phase reactant in cats. Serum levels increase upto 8-fold. In contrast to other assays for ceruloplasmin, the cat ceruloplasmin SPARCL assay (CER-SP-8) measures protein mass, not the oxidase activity. As with our other acute phase protein SPARCL kits, this assay requires a single no-wash, 30-min incubation. Refer to the SPARCL kits webpage for pricing.

Human LRG1 Antibodies – added October 18, 2017
We have just finished development of three human LRG1 antibodies, two  mouse monoclonals (LRG1-20-8G10 and LRG1-20-13H5) and a rabbit polyclonal (LRG1-20-RB). Dog Ceruloplasmin ELISA Kit – added September 21, 2017
We recently purified dog ceruloplasmin and made antibodies. These were used to develop the dog ceruloplasmin ELISA kit (catalog number CER-4). Ceruloplasmin is present normal dog serum at concentrations of ~0.1 mg/ml. Levels increase 3-fold or more during the acute phase response. Cat CRP SPARCL™ Kit – added August 29, 2017
The cat CRP SPARCL kit (CRP-SP-8) allows rapid measurement of CRP in cat serum and plasma. CRP is generally considered to be a poor acute phase protein in cats. In studies at Life Diagnostics however, we found CRP levels ranging from 42 ug/ml in serum from healthy cats to 360 ug/ml in serum from sick cats. Dog IgE SPARCL™ Kit – added July 18, 2017
The dog IgE SPARCL kit (IGE-SP-4) uses two of our recently developed monoclonal antibodies (see below). It allows rapid measurement of IgE levels in dog serum and plasma. Pricing can be found on the SPARCL kits page.

Anti-dog IgE monoclonals – added July 17, 2017
We have recently developed two anti-dog IgE monoclonal antibodies: IGE-4-4D1 and IgE-4-13H5. They can be used as a pair to develop ELISA and SPARCL assays for measurement of IgE in dog serum and plasma. The antibodies were developed by Life Diagnostics. We purify them under gentle conditions from serum-free media, avoiding low-pH elution steps that often partially denature antibodies. They are priced at $500/mg.

Monkey Leucine-rich alpha-2-glycoprotein-1 (LRG1) SPARCL™ Kit – added July 10, 2017
LRG1 is a positive acute phase protein that is elevated in human serum during inflammation and infection. It may also be a positive acute phase reactant in monkeys. Cat Serum Amyloid A (SAA)  SPARCL™ Kit – added April 25, 2017
SAA is a positive acute phase protein in cats. Its serum concentrations can increase >50 fold during inflammation and disease.  Similar to CRP in dogs, SAA serves as an excellent acute phase biomarker in cats (CRP is not an acute phase reactant in cats). Human Serum Amyloid A (SAA) – added October 22, 2016
Using our recently developed human SAA SPARCL™ kit we screened 90 human acute phase fluid samples and identified units with sufficiently high SAA levels to allow purification. The SAA (catalog# 9006) is highly purified, migrating as a single ~12 kDa band on SDS PAGE. It can be used in western blots, for antibody production, or to prepare assay controls and calibrators. Monkey SAA SPARCL™ Kit – added October 19, 2016
The kit allows rapid measurement of SAA in monkey serum and plasma (SAA-SP-3). Because SAA binds strongly to lipoproteins that interfere with SAA measurement, samples must be briefly treated with dissociation buffer (provided with the kit) prior to assay. SPARCL™ Kits – added May 12, 2016
Since November, 2015 the staff at LDI have been busy developing SPARCL™ versions of our ELISA kits. SPARCL™ technology (Spatial Proximity Analyte Reagent Capture) was developed by Lumigen Corp. We are very pleased to announce that Lumigen has granted Life Diagnostics exclusive rights to manufacture SPARCL versions of our ELISA kits. Thus far, we have made 35 kits. They are listed on our SPARCL kits webpage. More are in development.

SPARCL™ assays use two antibodies. One is conjugated to HRP, the second to acridan, a chemiluminescent substrate. When HRP and acridan conjugated antibodies both bind to their target antigen they are brought into close proximity. With the addition of hydrogen peroxide, HRP catalyzes oxidation of proximal acridan molecules causing a flash of chemiluminescence that is proportional to antigen concentration. Acridan conjugated antibodies distant from HRP produce no signal. The technology allows  development of rapid, no-wash, homogeneous immunoassays.

In a typical SPARCL™ assay a mixture of acridan and HRP conjugates is first pipetted into wells of a white 96-well SPARCL™ plate. Diluted samples and standards are then added. After a 30-minute incubation the plate is placed into a luminometer. Luminescence is measured after injection of trigger solution containing hydrogen peroxide, into each well. Assuming 15-minutes preparation and 5-minutes analysis time, the entire assay is complete within 50-minutes. This contrasts with ELISA’s that take 2 – 4+ hours, require 2-4 incubations and 1-3 wash steps.

SPARCL assays require a luminometer capable of simultaneous injection (only one injector is required) and measurement of luminescence. A list of suitable luminometers may be found here.

I have been developing ELISA’s since 2003 and have used them extensively for many years prior to that. I spent the last few months developing our SPARCL™ kits. I am very impressed with their simplicity and speed compared to ELISA.
